Preparation:
1.1, leaching: adding 8 times of 50-70% ethanol to the collected pine bark, and extracting the pine bark in 3 times.
1.2. Ethanol recovery: Evaporate the extract in vacuum, recover ethanol, add water to the extract and cool it for later use.
1.3. Centrifugation: The extract is subjected to three-stage centrifugation, among which, the first-stage centrifugation is mainly to remove large particle impurities in the extract, the second-stage centrifugation is mainly to remove small particle impurities in the extract, and the third-stage centrifugation is mainly to remove the extract The tiny particle impurities in the concentrated liquid can be effectively removed by three-stage centrifugation to remove solid impurities and water-insoluble particulate matter in the concentrated liquid, preventing the concentrated liquid from polluting the chromatographic column and making it difficult to regenerate.
2. Column desorption
2.1. The first pass
Pass the centrifuged extract through a macroporous resin chromatography column with a diameter-to-height ratio of 1:10. The macroporous resin chromatography column needs to be cleaned in advance to ensure cleanliness and resin regeneration. First wash with alkali, and then rinse with purified water to neutrality, then pickled with acid until the pH value ~ 4, and then rinsed with purified water to neutrality, after the extract was finished, washed through the column with purified water until the effluent was clear, and removed in the macroporous resin chromatography column Then use 5% to 15% ethanol, such as 5%, 10%, and 15% ethanol, to rinse in batches until the eluate is clear, so as to remove the molecular weight and macropores in the macroporous resin chromatography column. For substances with incompatible resin pore diameters, 70% ethanol is used to desorb the active ingredients in the macroporous resin chromatography column. After the desorption is completed, the desorption liquid is collected, and the ethanol is recovered under reduced pressure.
2.2. The second column pass
Pass the desorbed liquid through the column for the first time through a macroporous resin chromatography column with a diameter-to-height ratio of 1:30, that is, a macroporous resin chromatography column with a diameter-to-height ratio of 1:30, that is, a pore diameter of 500mm and a height of 15m Macroporous resin chromatography column, the macroporous resin chromatography column needs to be cleaned in advance to ensure that it is clean, that is, it has been regenerated. After setting out, wash the column with purified water until the effluent is clear, and then use 30% and 50% , 70% ethanol rinse three times (30% once, 50% once, 70% once), collect the desorption solution respectively, reclaim ethanol under reduced pressure, after being concentrated into a thick paste, vacuum drying promptly gets the pine bark extract. The macroporous resin chromatography column is very clean (without too many impurities) after the extract is passed through the column for the first time, so it can be cleaned with 95% ethanol to realize resin regeneration.
